1. Enable Desktop Effects -> Mouse Click Animation
2. Start clicking

Enabling the effect in System Settings simply allows this mode to be turned on and off, but you still need to do that manually. The default keyboard shortcut to do so is Meta+Shift+8. You can see this or change it in the applet's settings, accessible by clicking on the Configure button on the right side of its list item in System Settings.
