Old version of the title:
" Krita version 4.3.0, file size 3.4 GiB so far; (I just hit 100 layers, but there's still 4.5 GiB space LEFT, so the document can hold 7.9 GiB total). For some reason the program won't draw: the cursor just stays put and doesn't stroke. It is NOT my tablet"

Try going to Configure Krita -> Display, change to the other option, OpenGL or ANGLE, and restart Krita. Check if it works. Please come back and change the status to RESOLVED/FIXED if it does.

Also for the future, please in case of errors that you suspect cannot affect all people - like in this case, if drawing was unable in Krita we wouldn't release a new version, right? - or of it's something related to input, tablets, etc. please first go to a user support forum like krita-artists.org. It is much more user-oriented and user-friendly than this site, which is just a development-oriented website with a kind of a TODO list for developers. On krita-artists.org forum there will be people to help you with issues like that without the need to inform the developers team.
