SUMMARY
Can't 

STEPS TO REPRODUCE
1. Open Spectacle
2. Click on configure

OBSERVED RESULT
There is no After taking a screenshot section.

EXPECTED RESULT
There should be a after taking a screenshot section which should allow me to select whether I would lie to save to default location or save directly to clipboard.

You need Spectacle 19.12.0+
