When changing settings in the annotation editor of spectacle it would be nice, if the changed settings (e.g. used color, rectangle without fill) are saved and reused for the next annotation process.

On my system the settings are remembered (Version 22.03.70).

For me (Version 21.12.0) that doesn't work - I have to reapply the settings every time Spectacle is reopened.

Where did you get version 22.03.70 from?

I'm running KDE Unstable compiled from git master. It seems that this is implemented about a month ago in MR https://invent.kde.org/graphics/spectacle/-/merge_requests/105, so if you don't already have it working, it should be coming in the future when your distribution updates the version.
