In annotation mode when I select "No effect" in any shape or text tool the shadow is always applied and can't be removed.

This feature comes from an upstream library. Can you re-report it at https://github.com/ksnip/kImageAnnotator/issues/?

To change items shadow we need to access annotator settings which is not accessible from Spectacle.
